摘要:紫红曲(Monascus purpureus)是将真菌接种于蒸熟大米或谷物上发酵而成的一种紫红色米曲,具有消食、活血、健脾燥胃之功效,它可药食两用,紫红曲是红曲属中2个可食用菌种之一。红曲在我国的应用历史源远流长,不少史书中都对其有着详细记载。近年来,随着科学事业的不断发展,大量的科研资料表明,红曲具有降血脂、降血压、降血糖等多种药理活性。
本论文通过利用包括正、反相常压色谱、正、反相常半制备型中压色谱和制备型HPLC以及凝胶色谱、薄层色谱等色谱方法,对红曲中组分L进行系统的分离纯化,从中得到一个化合物。
再利用1D-NMR波谱技术进行结构鉴定,并与已知文献对照后确定该化合物的结构为胡萝卜苷,该化合物为首次从红曲中发现。

Abstract:Monascus purpureus is a purple rice-koji of cooked rice fermented by Monascus purpureus. It has the functions of digestion, blood circulation, invigorating the spleen and stomach are dry, and can be used in medicine and food. Monascus purpureus is one of two edible fungus of Monascus. Monascus application in China has a long history, many books have detailed records. In recent years, with the continuous development of science, a lot of research data shows, Monascus has the functions of reducing blood fat, lowering blood pressure, lowering blood sugar and so on many kinds of pharmacological activities.
In this paper, a compound was isotated by using positive phase and reverse phase nomal pressure chromatography, positive phase and reverse phase semi-preparative medium pressure chromatography, preparative HPLC chromatography, gel chromatography and thin-layer chromatography methods.
Then its structure was identified by 1D-NMR spectroscopy and compared with the previous literatures. It was elucidated as Daucosterol. This compound was found for the first time from monascus.

本论文是应用各种分离纯化技术,包括正、反相常压色谱和制备型HPLC以及凝胶、薄层等色谱方法,对红曲中组分L进行系统的分离纯化。首先使用正相硅胶柱,用氯仿:甲醇溶剂系统进行梯度洗脱,之后又利用低压Flash进行了进一步的分离纯化,经薄层板检测,得到了一个较纯净的主斑点。最后用高压液相进行制备,在制备过程中,需要注意几点:由于样品溶解性不好,所以每次进针时,必须超声将样品溶解成透明状态时才能进针;我们分离得到的样品量比较大,所以在制备的过程中,需要花费很长时间;再则,我们用的是制备型自动接收液相,流速比较快,所以每天要配置很多流动相,以及还要将废液回收利用,这样工作量是很大的,需要很大的体力和耐力。经过相当长一段时间制备后,我们最终分离得到了一个单体化合物。然后利用核磁共振技术进行结构鉴定,并与已知文献对照后确定了该化合物的结构为胡萝卜苷,该化合物为首次从红曲中发现。
