摘要:本文分别以菌伞、菌柄的水提粗多糖作为对象,测定了各自的自由基清除率,并就姬菇纯多糖对过氧化氢诱导小鼠肝损伤的保护作用进行了研究。结果表明姬菇多糖具有较好的还原力和自由基清除能力,尤其是清除羟自由基、超氧阴离子自由基的能力较强,其抗氧化能力与多糖质量浓度有一定的量效关系。此外姬菇纯多糖还能显著减少小鼠肝脏中MDA含量。

Abstract:This paper take aqueous polysaccharide from the cap and stipeof Pleurotus cornucopiae and polysaccharides which is removed protein as objects, determined their in vitro radical clearance rate and the protective effects of pure polysaccharide to the hydrogen peroxide-induced liver injury in mice. The results show that Pleurotus cornucopiaepolysaccharides has good reducing power and free radical scavenging ability , especially in hydroxyl radical and superoxide anion radical. The antioxidant activity of polysaccharide is associated with its concentration. In addition ,Pleurotus cornucopiae pure polysaccharides can also significantly reduce MDA content in livers of mice by reducing the occurrence of lipid peroxidation.

通过姬菇多糖体外抗氧化试验发现,其对羟自由基、DPPH自由基、超氧阴离子自由基具有较好的清除能力并表现出一定的量效关系。姬菇整株粗多糖对自由基清除能力大小依次为:超氧阴离子自由基>羟自由基>DPPH自由基,其EC50分别是1.15mg/mL、2.16 mg/mL、6.49 mg/mL。
组织匀浆在温育条件下可以发生自氧化反应而产生自由基,这些自由基能通过攻击生物膜中的不饱和脂肪酸引发脂质过氧化反应,形成脂质过氧化物。这类物质在细胞中积累,能干扰细胞定位,引起蛋白质、核酸等生物大分子的结构变化,使细胞收到损害,甚至死亡。因此,测试MDA的量不仅可以反映出组织内脂质过氧化的程度,而且还能间接反映出细胞损伤的程度。
